Off-target interaction

To evaluate binding specificity with the Attana cell-based biosensor provides crucial information about the interaction to be studied such as detrimental off-target effects. This can easily be observed and characterized by using Attana’s unique “one-stop-shop" system where the the binding events can be determined from both a biochemical and cell-based assay.

Performing cell-based assays in the biologically relevant environment offered by the Attana Cell 200 biosensor makes it easier to predict how candidates will perform in situ early in the discovery process generating a concentrated downstream focus for only the truly relevant candidates. To determine if candidates bind with high specificity or display detrimental off-target interactions is important to obtain the full binding profile of the lead to be studied in order to recognize the successful candidates and eliminate those of poor quality.

Off-target interactions are binding events of the candidate to other components on the cell surface than the actual target site. In the body these interactions will not only affect the efficacy of a treatment but can also give undesirable side effects. These unspecific interactions are often of low affinity and easily missed out in classical endpoint cell assays and falsely brought into the next step in the discovery process and then finally fail in clinical trials as a candidate with “low efficacy”.  

With Attana Cell 200 all kinds of macromolecular interactions to binding partners at the cell surface can easily be analyzed such as detrimental off-target interactions, surface target accessibility, avidity and drugability. Using Attana’s proprietary cell specific COP-1 surface coating, cells will be growing in a tissue culture flask-like environment directly on the sensor chip, displaying their true biological surface expression profile and morphology. The sample to be analyzed can be crude or even serum and is injected in a continuous flow over the cell containing sensor surfaces. Binding events to the cells are monitored in real-time and any off-target interaction will be discovered. Validation of the interaction is easily done with the sophisticated software option called TraceDrawer where several advanced binding models are included.
